The assay was carried out according to the method of Wardlaw and Willenbrink (1994) with slight modifications. The reaction mixture (110 µl) contained 50 mM Hepes-NaOH (pH 7.5), 100 mM fructose-6-phosphate(for SPS) or fructose(for SS), 100 mM UDPglucose, and 50 mM MgCl2. The reaction was started by adding 50 µl of crude enzyme extract. After incubating at 30C for 30 min, the reaction was stopped by boiling for 1 min. After adding 200 µl of 2 M NaOH, the solution was incubated for 10 min at 100C. After cooling, 2 ml of 30% (v/v) HCl and 1 ml of 0.1% (w/v) resorcin were added and the solution incubated for 10 min at 80C. Finally, sucrose content was determined by measuring the absorbance at 480 nm with a spectrophotometer.
